Reassociation kinetics of three satellite components of calf thymus DNA.

چکیده
Using absorption measurements the reassociation kinetics of three satellite DNA components isolated from calf thymus was studied under various conditions. A different method using CsC1 density gradient determinations particularly suited for kinetic analysis of mixtures was also used and shown to give similar results. Reassociation rate constants were corrected for mismatching during strand reassociation using data obtained by kinetic analysis of fractions of the 1.714 g/cm-3 satellite component. The values of corrected as well as uncorrected complexities were calculated and compared with results of other methods. They were shown to be compatible with the concept of sequence repetition at various levels.

It has been proved that carcinogens bind to macromolecules of the target tissues [ 1,2] and that a positive correlation exists between the extent of binding of hydrocarbons to DNA and their carcinogenic properties [3] . The binding of hydrocarbons is coupled with their metabolic activation [4,5]. The ultimate metabolite of benzo(a)pyrene (BP) as suggested by Boyland [6] has been isolated as a K...
متن کاملSpectroscopic study on the interaction of three water-soluble porphyrins with calf thymus DNA
Porophyrins and their metal derivatives are strong DNA binders with association constant of to . Some of these compounds have been used for the radiations sensitization therapy of cancer and are targeted to interact with cellular DNA. Binding of porphyrins to DNA changes the expectral and other physic chemical characteristic of porphyrins, The mode of binding can be extracted from inspection ...
متن کاملDistribution of 5-methylcytosine in the DNA of somatic and germline cells from bovine tissues.
Genomic DNA of calf thymus contains 1.5 times as much 5-methylcytosine as similar sperm DNA, but the major EcoRI repeat fragment from satellite I of thymus contains ten times as much 5-methylcytosine as the corresponding fragment from sperm DNA. Restriction enzyme analyses of the total DNA and the satellite I fragment show that three HpaII sites in the fragment are completely unmethylated in sp...
متن کاملReassociation Kinetics and Cytophotometric Characterization of Peanut (Arachis hypogaea L.) DNA.
The base composition of peanut (var. NC-17) DNA determined from thermal denaturation profiles showed an average guanine plus cystosine content of 34% which was in close approximation to 36% guanine plus cytosine calculated from the buoyant density. Buoyant density also indicated the absence of satellite DNA. The genome size, 2.0 x 10(9) base pairs, as determined by reassociation kinetics of the...
